Plantlet Production throught Callus Culture in Allium sativum L.



Abstract
Organogenic abilities and growth patterns of callus obtained from shoot tip culture in Allium sativum as affected by several kinds of plant growth regulators and additives and subculture times were investigated and the changes of structure during culture were also examined by scanning electron and light microscope.
It was possible to produce friable and compact callus from shoot tip culture by manipulating concentration of growth regulators and casamino acid added to media. Embryo-like tissues from compact callus developed roots. Shoot formation and callus growth were better with friable callus than with compact ones under light condition. Friable callus was obtained with media containing 0.1 §·/L 2iP, 0.5 §·/L NAA and 0.1 §·/L 2.4-D, was optimum for subculture. Observation with SEM and light microscope revealed development of adventitious buds originating from inside or surface of callus into shoots. Addetion of 10 §·/L BA was most effective on shoot formation. Organogenic ability was reduced from 60% to 10% after one year of subculture.
